    Managing the acidity of abandoned water filled coal mining voids in Collie (Western Australia) using organic matter

    The effectiveness of using organic matter additions to increase pH in abandoned water filled coal mining voids in Collie was investigated. Previous studies have demonstrated that passing acidic drainage through organic matter increased the waters pH. Laboratory trials using intact sediment cores (collected from Ewington, Collie, Western Australia) were used to assess the effects of additions of hay, manure and mulched vegetation on pH, nutrients an sulphate levels. In a series of experiments the effect of different quantities of organic material, sulphate reducing bacterial inoculations and alkalinity generation were measured. A subsequent field experiment was conducted to examine the impact that manure and mulch had on mine void water in 18 in situ ponds constructed adjacent to Ewington. It was concluded from the laboratory experiments that mulch and manure treatments were significantly better than hay as an organic matter addition for increasing the pH of acidified mine void water at Ewington. The manure produced the greatest increase in pH, although it contributed less to alkalinity than mulch; suggesting mulch bas a greater ability to release carbonates than did manure. The manure and mulch additions increased the pH in the laboratory and field experiment by 0.5-1.5 pH units. This increase was sustained for most of the 21 week field experiment. An increase in pH in the control ponds was recorded over the last 8 weeks of the experiment which was probably due to the inflow of more alkaline groundwater through the substratum as a consequence of the winter rainfall. pH values measured in the experimental ponds 15 months after the commencement of the project indicated that the ponds treated with manure maintained significantly higher pH levels than either the mulch or the control ponds. There was no difference between the pH values for the ponds treated with mulch and the controls suggesting that manure not only provided a greater increase in pH but also over a longer period. The addition of organic mailer also resulted in an increase in gilvin in the laboratory experiments. Low sulphate and sulphide levels were also recorded in all experiments before and after the introduction of organic materials into mine void water and ponds adjacent to Ewington indicating that sulphate reducing bacterial activity was not the cause for the increase in pH as occurred in other situations, but rather the addition of alkaline organic matter caused the increase in pH. The addition of manure organic material was associated with an increase in the ortho-phosphate levels, resulting in an increase in chlorophyll a concentrations. It is believed to be the first stages of succession processes leading to the establishment of a biologically active wetland system. During this process the emerging ecosystem neutralises the acidic content of the water

    Retrospective application of remote sensing to the Tasmanian lakeland

    This thesis describes the research undertaken for a degree of Master of Science in a retrospective study of airborne remotely sensed data registered in 1990 and 1993, and field captured data of aquatic humus concentrations for ~ 45 lakes in Tasmania. The aim was to investigate and describe the relationship between the remotely sensed data and the field data and to test the hypothesis that the remotely sensed data would establish further evidence of a limnological corridor of change running north-west to south- east. The airborne remotely sensed data consisted of data captured by the CSIRO Ocean Colour Scanner (OCS) and a newly developed Canadian scanner, a compact airborne spectrographic imager (CASI). The thesis investigates the relationship between the two kinds of data sources. The remotely sensed data was collected with the OCS scanner in 1990 (during one day) and with both the OCS and the CASI in 1993 (during three days). The OCS scanner registers data in 9 wavelength bands between 380 nm and 960 nm with a 10-20 nm bandwidth, and the CASI in 288 wavelength bands between 379.57 nm and 893.5 nm (ie. spectral mode) with a spectral resolution of 2.5 nm. The remotely sensed data were extracted from the original tapes with the help of the CSIRO and supplied software and digital sample areas (band value means) for each lake were subsequently extracted for data manipulation and statistical analysis. Field data was captured concurrently with the remotely sensed data in 1993 by lake hopping using a light aircraft with floats. The field data used for analysis with the remotely sensed data were the laboratory determined g440 values from the 1993 water samples collated with g440 values determined from earlier years. No spectro-radiometric data of the lakes, data of incoming irradiance or ancillary climatic data were captured during the remote sensing missions. The sections of the background chapter in the thesis provide a background to the research both in regards to remote sensing of water quality and the relationship between remotely sensed spectral data and water quality parameters, as well as a description of the Tasmanian lakes flown. The lakes were divided into four groups based on results from previous studies and optical parameters, especially aquatic humus concentrations as measured from field captured data. The four groups consist of the ‘green” clear water lakes mostly situated on the Central Plateau, the ‘brown” highly dystrophic lakes in western Tasmania, the ‘corridor” lakes situated along a corridor of change lying approximately between the two lines denoting the Jurassic edge and 1200 mm isohyet, and the ‘eastern, turbid” lakes make up the fourth group. The analytical part of the research work was mostly concerned with manipulating and analysing the CASI data because of its higher spectral resolution. The research explores methods to apply corrections to this data to reduce the disturbing effects of varying illumination and atmospheric conditions. Three different methods were attempted. In the first method two different standardisation formulas are applied to the data as well as ‘day correction” factors calculated from data from one of the lakes, Lake Rolleston, which had data captured for all three days of the remote sensing operations. The standardisation formulas were also applied to the OCS data. In second method an attempt to reduce the effects of the atmosphere was performed using spectro-radiometric captured in 1988 for one of the lakes flown, Great Lake. All the lake sample data were time normalised using general irradiance data obtained from the University of Tasmania and the sky portion as calculated from Great Lake upwelling irradiance data was then subtracted. The last method involved using two different band ratios to eliminate atmospheric effects. Statistical analysis was applied to the data resulting from the three methods to try to describe the relationship between the remotely sensed data and the field captured data. Discriminant analysis, cluster analysis and factor analysis using principal component analysis (pea) were applied to the remotely sensed data and the field data. The factor scores resulting from the pca were regressed against the field collated data of g440 as were the values resulting from last method. The results from the statistical analysis of the data from the first method show that the lakes group well (100%) against the predetermined groups using discriminant analysis applied to the remotely sensed CASI data. Most variance in the data are contained in the first factor resulting from pca regardless of data manipulation method. Regression of the factor scores against g440 field data show a strong non- linear relationship and a one-sided linear regression test is therefore considered an inappropriate analysis method to describe the dataset relationships. The research has shown that with the available data, correction and analysis methods, and within the scope of the Masters study, it was not possible to establish the relationships between the remotely sensed data and the field measured parameters as hoped. The main reason for this was the failure to retrieve remotely sensed lake signatures adequately corrected for atmospheric noise for comparison with the field data. This in turn is a result of the lack of detailed ancillary information needed to apply available established methods for noise reduction - to apply these methods we require field spectroradiometric measurements and environmental information of the varying conditions both within the study area and within the time frame of capture of the remotely sensed data

    Functional Reconstitution of a Tunable E3-Dependent Sumoylation Pathway in Escherichia coli

    SUMO (small ubiquitin-related modifier) is a reversible post-translational protein modifier that alters the localization, activity, or stability of proteins to which it is attached. Many enzymes participate in regulated SUMO-conjugation and SUMO-deconjugation pathways. Hundreds of SUMO targets are currently known, with the majority being nuclear proteins. However, the dynamic and reversible nature of this modification and the large number of natively sumoylated proteins in eukaryotic proteomes makes molecular dissection of sumoylation in eukaryotic cells challenging. Here, we have reconstituted a complete mammalian SUMO-conjugation cascade in Escherichia coli cells that involves a functional SUMO E3 ligase, which effectively biases the sumoylation of both native and engineered substrate proteins. Our sumo-engineered E. coli cells have several advantages including efficient protein conjugation and physiologically relevant sumoylation patterns. Overall, this system provides a rapid and controllable platform for studying the enzymology of the entire sumoylation cascade directly in living cells

    An Investigation of the Floor Attachment Assembly of a Large Transport Aircraft Seat Subjected to Crash Dynamic Loadings

    Accident investigations by the National Transportation Safety Board concluded that the passenger seat and restraint systems found in today\u27s aircraft used by the airlines are inadequate to protect the occupant in airplane crashes. Computer models were developed of the seat-track assembly and finite element analyses were performed subjected to crash dynamic loadings. The seat-track interface was modeled with gap elements to accurately model the geometric interaction between the track and seat-track fittings. Results showed large deformations and stresses above yielding in both the seat and track when subjected to accelerations prescribed for certification under the Federal Aviation Administration (FAA) regulations

    Lidar remote sensing and co-operative observations: Processing methods and aerosol radiative transfer

    This Ph.D. thesis focuses on: (i) the design and integration of a polarimetric channel for the multi-spectral Raman lidar station of the Universitat Politècnica de Catalunya (UPC), Remote Sensing, Antennas, Microwaves and Superconductivity Group (CommSensLab), (ii) the study of the temporal and spatial evolution of atmospheric aerosol optical, microphysical and radiative properties by means of active and passive remote sensing in the context of ACTRIS and Spanish National projects, and (iii) rainfall rate retrieval by means of a vertically-pointed ceilometer in the context of the Verification of the Origins of Rotation in Tornadoes Experiment Southeast (VORTEX-SE). The first goal of this Ph.D., tested on the UPC multi-spectral Raman lidar station, consists of developing a secondary optical receiving chain, installed next to the laser source. The secondary telescope, mounted in the optical chain, allows retrieving the cross-polarized return signal separately from the total-power signal, avoiding the need of a very precise characterization of the crosstalk parameters of the beam-splitters. The first experimental results, corresponding to a collection of atmospheric conditions over the city of Barcelona, are also presented. The second goal of this Ph.D. deals first with the GAME (Global Atmospheric Model) code, necessary to retrieve the aerosol radiative properties. The radiative fluxes estimated in the short-wave and long-wave spectral ranges at the bottom and the top of the atmosphere by GAME are compared to the ones retrieved by a different radiative transfer model, namely Two-Stream, in order to know the importance of the spectral parameterization of a radiative transfer code. Then, GAME code, in both configurations, is fed by means of three different datasets to evaluate the parameterization of the vertically-resolved properties and to assess the uncertainty of GAME when is tuned with input parameters from different sources. Afterwards, an evaluation of the seasonal variability of the aerosol background optical and radiative properties in the Western Mediterranean Basin (WMB) is performed by means of AERONET (Aerosol Robotic Network) sun-photometers data from two background sites, Ersa (Corsica Island, France) and Palma de Mallorca (Mallorca Island, Spain). In addition, in order to detect possible northeast-southwest gradients in the aerosol properties, a third site located at Abolrán (Alborán Island, Spain) is considered. Finally, during 15-24 June 2013 a moderate Saharan dust multi-intrusion was detected by some EARLINET/ACTRIS (Granada, Barcelona, Naples, Potenza and Serra la Nave (Italy)) and ADRIMED/ChArMEx (Cap d’en Font, (Minorca Island, Spain) and Ersa) stations. This Ph.D. uses this event to study the spatio-temporal evolution of the mineral dust properties, since the lidar stations were supported during the multi-intrusion by collocated AERONET sun-photometers and the Falcon 20 aircraft. Also the GAME code is used to estimate the aerosol radiaite effect during the Saharan dust event. Besides, air- and space-borne lidar measurements, satellite images and back-trajectories are used to confirm the multi-intrusion aspect of the event. The last goal of this Ph.D. pursues estimation of the rain rate (RR) from ceilometer measurements. In VORTEX-SE, a Vaisala CL-31 ceilometer, a S-band radar, and a disdrometer were deployed in Alabama during March-April 2016. First, rain-extinction coefficients from ceilometer attenuated backscatter measurements are derived by means of a modified form of the well-known slope-method. These coefficients are compared with the RRs measured by a collocated S-band radar and a disdrometer in order to get the RR-to-extinction models. Advanced covariance-weighted techniques are used to best assess and validate the estimated models. These models can be used to estimate the RR from the ceilometer in similar situations without need to have a collocated cooperative instrument permanently deployed.Este Ph.D. se centra en: (i) en el diseño e integración de un canal polarimétrico para la estación lidar multi espectral del grupo de teledetección, antenas, microondas y superconductividad (CommSensLab) de la Universitat Politècnica de Catalunya (UPC), (ii) en el estudio de la evolución temporal y espacial de las propiedades ópticas, microfísicas y radiativas de los aerosoles por medio de teledetección activa y pasiva en el contexto de ACTRIS y proyectos estatales, y (iii) en la recuperación de intensidad de lluvia por medio de un ceilómetro en configuración vertical en el contexto del proyecto Verification of the Origins of Rotation in Tornadoes Experiment Southeast (VORTEX-SE). El primer objetivo, realizado en la estación lidar de UPC, consiste en el desarrollo de una cadena óptica secundaria instalada junto al láser. El telescopio secundario, montado en la cadena óptica, permite recuperar la componente cross-polarized de la señal total por separado, evitando la necesidad de conocer con precisión los parámetros de los beam-splitters. Se presentan también los primeros resultados obtenidos en Barcelona durante diferentes situaciones atmosféricas. El segundo objetivo de este Ph.D. se centra en el código GAME (Global Atmospheric Model), necesario para recuperar las propiedades radiativas de los aerosoles. Los flujos radiativos estimados tanto en onda larga como en onda corta en la base y en la parte superior de la atmósfera son comparados con los estimados por otro código de transferencia radiativa, Two-Stream, para conocer la importancia de la parametrización espectral. Después, el código GAME es alimentado con 3 bases de datos diferentes para evaluar la parametrización de las propiedades resueltas en altura y conocer la incertidumbre de GAME cuando es alimentado con parámetros con diferentes orígenes. Por otro lado, se presenta una evaluación de la variabilidad estacional de las propiedades ópticas y radiativas del aerosol de fondo en la cuenca oeste mediterránea (WMB) realizada con datos de fotómetros solares de la red AERONET (Aerosol Robotic Network) situados en dos puntos considerados libres de contaminación: Ersa (isla de Córcega, Francia) y Palma de Mallorca. Además, para detectar posibles gradientes noreste-suroeste en las propiedades delos aerosoles, se considera un tercer punto ubicado en la isla de Alborán. Por último, en este Ph.D. se aprovecha una multi intrusión moderada de polvo sahariano, detectada entre los días 15 y 24 de junio de 2013 por algunas estaciones EARLINET/ACTRIS (Granada, Barcelona, Nápoles, Potenza y Serra la Nave (Italia)) y ADRIMED/ChArMEx (Cap d'en Font (Menorca) y Ersa), para estudiar la evolución espacio-temporal de las propiedades del polvo mineral, ya que las estaciones lidar estaban apoyadas durante el evento por fotómetros solares pertenecientes a la red AERONET, situados junto a las estaciones lidar, y por vuelos del Falcon 20. GAME es usado para obtener también el efecto radiativo de los aerosoles durante el evento de polvo sahariano. Para confirmar el aspecto de multi intrusión se utilizan medidas lidar tomadas a bordo de aviones y satélites, imágenes satelitales y retro trayectorias. El último objetivo del Ph.D. persigue la estimación de la RR utilizando medidas de un ceilómetro. En VORTEX-SE, se desplegaron (Alabama, marzo-abril 2016) un ceilómetro Vaisala CL-31, un radar de banda S y un disdrómetro. Se han estimado los coeficientes de extinción debida a la lluvia a partir del retorno atenuado medido por el ceilómetro, utilizando una versión modificada del método de la pendiente. Estos coeficientes se comparan con las intensidades de lluvia (RR) estimadas con el radar y el disdrómetro para obtener modelos de RR-extinción. Para validarlos se utilizan técnicas avanzadas de covarianza ponderada. Dichos modelos pueden usarse para estimar la RR con un ceilómetro, en situaciones similares, sin necesidad de tener desplegado permanentemente un instrumento cooperativo.Postprint (published version

    Clarifying the waters: A critical analysis of turbidity and its role in environmental monitoring in New Zealand

    Turbidity is widely-used as a water quality indicator to infer the mass of suspended sediment (SS) transported through riverine systems, and is inexpensive, readily available, and can be easily deployed to record continuous measurements. However, using turbidity as a surrogate for SS concentration (SSC) is frequently confounded by the composition of riverine suspended material, and the particle size and shape of sediments. Presented as a series of research papers, this thesis provides a distinctly Southern Hemisphere perspective of sediment, organic matter, organic carbon, and turbidity across southern New Zealand, and offers a critical reflection on the role of turbidity in environment monitoring. The suspended material of southern New Zealand rivers under base flow was mostly comprised of inorganic suspended material, although the total composition also includes variable amounts of organic material (<10%, and up to 50 – 80%). The proportion of organic to inorganic particulate material in southern New Zealand is significant under certain land covers, and contributes a significant proportion of particulate organic carbon flux to the Southern Oceans (0.04 – 2.7 t km-1 a-1). In addition, different catchment morphologies and lithologies have a propensity to discharge different particle size distributions of inorganic sediment. These differences in particle sizes are most likely a function of underlying lithology from in channel attrition. This thesis has also identified that specific turbidity (turbidity normalised to mass concentration of particulates) is a potentially effective metric to indicate a ‘non-standard’ light attenuation response (that is, greater turbidity per mass of SS). Specific turbidity is shown through both in-field measurements and laboratory experiments to be affected by organic composition (particulate and dissolved) and the particle size distribution. Multiple regression analysis of catchment characteristics show that suspended particulate material composition and particle size are linked to discharge and flow behaviour, landcover, and lithology. The application of these findings is applied to monitoring SSC with turbidity across New Zealand by examining the specific turbidity for 77-monitoring stations. It is evident that organic composition and particle size have a notable effect on SS-turbidity ratings across New Zealand, which has limitations for the comparability, and relevance of SS data derived from turbidity when used as a regulatory tool. This thesis shows that although turbidity is pragmatic, its use in SS monitoring is problematic

    Trace metal speciation in the Pieman River catchment, western Tasmania

    The Pieman River catchment has seen continuous mining of economic deposits of gold, silver, lead, copper, zinc and tin since the 1870’s. Tributaries of this river which receive mining effluent, either directly or from acid mine drainage (AMID), have total metal concentrations considerably above background levels and are of regulatory concern. The lower Pieman River is however classified as a State Reserve in which recreational fishing and tourism are the major activities. It is therefore important that water entering the lower Pieman River from upstream hydroelectric impoundments is of high quality. Metals in natural waters exist in a variety of dissolved, colloidal and particulate forms. The bioavailability and hence toxicity of heavy metal pollutants is very dependant on their physico form. Knowledge of the speciation of a metal in natural aquatic environments is therefore necessary for understanding its geochemical behaviour and biological availability. Complexation of metal ions by natural ligands in aquatic systems is believed to play a significant role in controlling their chemical speciation. This study has investigated temporal and spatial variation in complexation of metal ions in the Pieman River. The influence of pH, temperature, organic matter, salinity, ionic strength and time has been investigated in a series of field studies and in laboratory-based experiments which simulated natural and anthropogenic disturbances. Labile metals were measured using two techniques in various freshwater and estuarine environments. Diffusive gradients in thin-films (DGT) allowed in situ measurement of solution speciation whilst differential pulse anodic stripping voltammetry (DPASV) was used to measure labile metal species in water samples collected from the catchment. Organic complexation was found to be a significant regulating mechanism for copper speciation and the copper-binding ligand concentration usually exceeded the total copper concentration in the river water. Complexation was highly dependent on pH and at the river-seawater interface was also regulated by salinity, probably as a result of competitive complexation by major ions in seawater (eg. Ca 2+ ions). Zinc complexation was also evident, however total zinc concentrations in the water column often far exceeded the potential binding capacity of available ligands. In addition to organic complexation, Zn speciation may also be associated with adsorption by flocculated or resuspended colloidal Mn and/or Fe oxyhydroxides. Metal ion complexation and hence speciation was found to be highly variable within the Pieman River catchment. This presents major difficulties for environmental managers, as it is therefore not possible to make catchment-wide assumptions about the bioavailability of these metals. These results emphasise the importance of site-specific sampling protocols and speciation testing, ideally incorporating continuous, in situ monitoring

    Energy efficient electric lighting for buildings in developed and developing countries

    As energy is a fundamental service for human development and economic growth, the demand for it is constantly on the rise worldwide. Lighting energy use makes a significant contribution to the total energy consumption of buildings. The use of energy efficiency measures can reduce this kind of energy consumption. The main objectives of this work were to review different aspects of lighting quality and energy efficiency and to test the existing technologies for efficient lighting. An additional aim of the work was to examine the new opportunities provided by LED technology in providing lighting in rural areas of developing countries and to compare LED lighting with existing fuel-based lighting. Three different lighting control systems in office rooms were compared for energy efficiency and the quality of lighting by means of measurements. The results of the measurements showed a significant potential for saving energy by the use of daylight-based dimming and occupancy control. The renovation of an auditorium with a new lighting installation resulted in higher illuminance levels and better colour rendering, while reducing energy consumption. This work also presents a calculation of lighting energy use in office rooms using two different calculation methods and discusses the different parameters used for the calculation. A comparison of the calculated values with the measured values confirmed the accuracy of the calculation methods. The work presents a study and evaluation of traditional pine stick lighting and new white LED-based lighting used in rural Nepali villages. The use of different renewable energy sources in combination with efficient lighting technology is found to be a realistic and sustainable option to provide clean and efficient lighting services in developing countries
